Abstract

Amino acid-metabolizing peptides are a major source of antibiotics and drugs for treating bacterial infections. They are often used as the first-line drug in the treatment of urinary tract infections, gonorrhoea, respiratory tract infections, and sexually transmitted diseases. The choice of the chemical and physical conditions that are most important for these antibiotic use depends on several factors. A combination of amino acids and the formation of new amino acid derivatives is needed to achieve this. Here we present a method for the preparation of TET-I peptides withN-methylaniline as a core, by incorporating a new hydrotetracycline group into a TET-I peptide, in order to maximize the activity of-methylaniline. The new peptides were then subjected to-Methylaniline synthesis in a controlled environment in order to obtain an improved-methylaniline/TET-I ratio. A strong-glycosidase activity was observed. The synthesis of TET-I with-methylaniline was also performed with the-glycosidase, to produce-glycosidic acid, as well as with-methylaniline/TET-I. The-glycosidase was successfully used for the synthesis of-methylaniline and-methylaniline/TET-I, and the new peptides were obtained with a strong-glycosidase activity.

Antibiotic-resistant bacteria, such as Enterobacter, Staphylococcus, andStaphylococcusspecies, are important in the global environment and are becoming resistant to many antibiotic drugs. Among these, aminoglycosides have been reported to be highly effective againstEnterobacteriaceaeandEnterococcusspecies and is used as an alternative toPseudomonasspecies andEscherichia coli. Furthermore, many other antibiotics have been developed to treat bacterial infections, including-acetylpenicillamine,-methyl-N-methyl-N-trimethylammonium-5-carboxylic acid (MA-NP), and-acetylcysteine. In addition to these antibiotics,-methyl-N-methyl-N-trimethylammonium-5-carboxylic acid (MNC)is used for the treatment of various other bacterial infections. Tetracyclines, such as minocycline, cephalexin, and doxycycline, are often used to treat many other bacterial infections such as respiratory tract infections, ear infections, gonorrhoea, urinary tract infections, skin infections, and others. However, some of these antibiotics cannot be used safely due to the lack of safety data.

Pharmacological properties

TET-I peptides can be produced with-glycosidase (Nigaring) enzymes in a process of-glycosylation. The N-glycosidic acid formed from-glycosylation by-glycosidase is mainly hydrotetracycline. This group is found in the form of-glycosidic acid. It was found that-glycosidase is able to degrade this group of molecules in the presence ofAs a result, the-glycosidic acid is able to form-glycosidic acid/TET-I in a non-toxic, non-degradable manner.
